Recovery of Injured Salmonella typhimurium with Hydrophobic Grid Membrane Filter

Abstract

|
To facilitate recovery of injured Salmonella typhimurium while providing selectivity of isolation of S. typhimurium from other bacteria in the safety assessment of the food sample, hydrophobic grid membrane filtration (HGMF) was used. Heat- or acid-injured S. typhimurium were filtered on HGMF and placed on Tryptic soy agar (TSA) plates for 6 hr at 37¡É. After resuscitation, the filters were transferred to Xylose lysine decarboxylase agar (XLD; selective medium) (HGMF/XLD). The selective agent in XLD will be diffused to HGMF to inhibit the growth of non-target microorganisms without effects on recovery of target microorganisms. The recovery rates of heat (55¡É for 10 min)- and acid (2% acetic acid for 1 min)-injured S. typhimurium by HGMF/XLD were compared to TSA and XLD. The heat- or acid-injured S. typhimurium grew and produced a typical S. typhimurium reaction (black color on HGMF/XLD) after 24 hr of incubation at 37¡É. No significant difference occurred for recovery of the injured S. typhimurium between TSA and HGMF/XLD (P>0.05), but higher numbers were recovered on TSA and HGMF/XLD than on XLD (P<0.05).
